I am a long time user of Thunderbird. I've been running it against my hosted servers since 2003 on Windows 7 and Mac OS.

I installed TB on Windows 10 this week, but was unable to connect to the server to read email.

I used the CMD line to telnet {mail.server name} pop3 and was able to connect with the username and password without any issues.

This machine has Microsoft Mail installed on it, which is confused with the reply address using the name of the mail server, instead of the User's name.

Is it possible that Windows 10 (really not your friend) is interfering with the Protocols to route them to Microsoft Mail if it's installed?

How do I fix this virus we call Windows 10?

exactly what is the error message you are receiving.

Please also post the account setting from the Troubleshooting information on the help menu. I don't want the email addresses, but server names and ports are necessary for testing purposes. As you say it is your site, who is the host. This can be the $64 question as they are not all created equal.

There is no error message. There is no connection from the computer to my server either. It's like there is something blocking the POP3 connection to the mail server.

Here's a point that I think was missed in my original post.

I tested the connectivity from the desktop to the server using the telnet command.

telnet <pop3 server name.com> pop3 user <username> pass <password>

This checked out perfectly.

When I tried to connect to the server via TB, the connection never reaches the pop3 server.

What could be interfering?

It is probably the firewall component in your anti virus... this is particularly common with Norton brand anti virus, but they are all rubbish with updates to some extent. They should have stayed with what they knew, but defended made them obsolete.
