It is much easier than that. For Thunderbird prior to v45 see http://kb.mozillazine.org/Setting_and_changing_email_passwords
As of Thunderbird v45 you can also directly edit account passwords Thunderbird has remembered. At the top right of the Thunderbird window, click the menu button > Options > Security > Passwords tab > Saved Passwords > Show Passwords. Right-click an entry to edit an existing password.

Please explain what you're trying to achieve in the first place.
If the password for the external e-mail account (i.e. gmail, hotmail) changes, there does not seem to be a way in Thunderbird to change the password for getting into the email account without deleting the email ?profile? and reentering the email address, password, and settings.
