For text in the messages themselves, go to Tools|Options|Display|Formatting→(Advanced button)
Set your font sizes there, in both "Latin" and "Other writing systems" to cover most of the bases.
Note the size settings are in pixels, not points, and so you'll need to set them to larger values. Start at 14 and go up.
For the general text in the Thunderbird list display, this add-on is useful:
