Thunderbird will not upgrade to 52.2.0
When I endeavor to update Thunderbird from 52.1.1 (32-bit) to 52.2.0, I get this message: "The update could not be installed. Please make sure there are no other copies of Thunderbird running on your computer, and then restart Thunderbird to try again." I have no other copies running (I checked, only have one profile in C:\Users\<name>\AppData\Roaming\Thunderbird\Profiles) but the problem persists. The error message now displays each time I start Thunderbird. Running app on Windows 10 Home, Version 1607 for x64-based Systems.

We've had a few examples of this recently.
Delete the Mozilla Thunderbird directory at %ProgramFiles(x86)% Reinstall Thunderbird. If it still fails, start Windows in safe mode and repeat.
This won't delete your Thunderbird data. But it's always wise to have periodic backups https://support.mozilla.org/en-US/kb/profiles-where-thunderbird-stores-user-data
1) 32 bit version in play on a x64 version of Home 1607? reasoning? 2) Copies is not referring to 'profiles' but whether there is an instance of Thunderbird presently running (opened). 3) Have you tried a standalone upgrade? 3a) Download the .exe ( https://ftp.mozilla.org/pub/thunderbird/releases/52.2.0/win32/en-US/Thunderbird%20Setup%2052.2.0.exe ) 3b) From the C:\Users\<Username>\Downloads\ folder right click > Open/Install on the downloaded .exe 4) As long as the files/folders in C:\Users\<username>\AppData\Roaming\Thunderbird\ are not messed with your previous settings should be auto populated on the upgraded version.
